Luciferase Complementation Assay (LCA) Detection Service

Luciferase Complementation Assay (LCA) Detection Service

High Sensitivity
Low Background
Real-time Monitoring
Non-invasive

Service Features

Compared to other protein interaction research technologies, the Luciferase Complementation Assay (LCA) using the tobacco transient expression system offers advantages of simplicity, sensitivity, reliability, efficiency, and low background. It can be easily scaled up for large-scale screening and validation of protein interactions.

Service Introduction

Luciferase Complementation Assay (LCA) is used to determine whether two proteins can interact. Two fragments of the luciferase protein are respectively linked to two different test proteins. If the two proteins can bind, the luciferase protein is complemented and catalyzes the substrate to emit fluorescence.

Service Advantages

  • High Sensitivity: LCA can detect protein interactions at picomolar levels, making it particularly suitable for studying low-abundance protein interactions.
  • Real-time Monitoring: Due to the rapid catalytic reaction of luciferase and strong bioluminescence signal, LCA enables real-time monitoring of protein interactions without cell fixation or lysis, reducing sample processing losses and variations.
  • Non-invasive Detection: LCA is performed in living cells without disrupting cell structure, allowing observation of protein interactions under physiological conditions and providing interaction information in a more authentic biological environment.
  • Wide Dynamic Range: LCA can cover a broad range of interaction strengths, from weak to strong interactions.

Customer Requirements

Gene synthesis by our company:

Provide the protein CDS sequence.

Customer provides template plasmid:

1. Customer provides vector/template: Need to provide vector MCS sequencing report/template sequencing results. If no sequencing report is available, additional sequencing fees will be charged, and sequencing will be performed by GeneCreate.

2. Customer provides bacterial culture: Culture volume should be ≥0.5mL. Glycerol stocks stored for over one year need to be activated before submission.

3. Customer provides plasmid: Plasmid quantity ≥5μg.

4. Both bacterial cultures and liquid plasmids require ice pack transportation.
