Experimental Principle
Firefly luciferase and Renilla luciferase catalyze the oxidation of luciferin or coelenterazine to form oxyluciferin or coelenteramide, respectively, producing bioluminescence in the process. The Double-Luciferase Reporter Assay Kit first detects the activity of the firefly luciferase reporter gene using luciferin as a substrate, then quenches this reaction while detecting the activity of the Renilla luciferase reporter gene using coelenterazine as a substrate. This system offers rapid detection, high sensitivity, broad detection range, and no interference from endogenous cellular activities.
Product Advantages
- This product demonstrates significantly higher detection sensitivity compared to similar products on the market, with stable chemiluminescent signals.
- The luminescence intensity is superior to other brands.
- Optimal firefly luciferase quenching effect, far exceeding other market brands, with minimal fluorescence residue that has almost no impact on Renilla luciferase reluminescence, improving test result accuracy and data reproducibility.
- Excellent stability, simple operation, stable readings, and fast detection speed.
Scope of Application
- miRNA Target Validation: Validate miRNA binding to target mRNA or lncRNA by constructing luciferase reporter vectors.
- Transcription Factor Regulation Verification: Clone transcription factor binding sites upstream of the firefly luciferase gene to verify regulatory effects by detecting changes in luciferase activity.
- Promoter Analysis: Clone promoter region sequences upstream of the firefly luciferase gene to analyze promoter activity and its changes under different conditions.
- Promoter SNP Analysis: Analyze relative activity of single nucleotide polymorphisms in promoter regions using the luciferase reporter system.
