Experimental Principle
GST Pull-Down assay is based on GST (glutathione-S-transferase) protein, which can bind to glutathione (GSH). GSH is immobilized on magnetic beads to form GSH-beads. When a known protein X is fused with GST, the resulting GST-X protein can bind to GSH-beads. If there is a protein Y that interacts with protein X in the system, a "bead-GSH-GST-X-Y" complex will form, allowing the interacting protein to be separated and detected.
Product Advantages
- High Specificity and Reliability: Strong affinity and stable binding between GST tag and glutathione beads significantly reduce non-specific background.
- Optimized Buffers: The kit provides upgraded and optimized buffers, saving time on experimental optimization.
- Cost-Effective: Rapid and convenient operation with costs significantly lower than in vivo experiments like Co-IP or yeast two-hybrid.
Scope of Application
- Protein Interaction Validation: Direct detection of protein-protein binding in vitro (e.g., kinase-substrate, receptor-ligand, transcription factor-DNA binding protein) for signaling pathway research or drug target development.
- Pathogen-Host Interaction Studies: Identification of interactions between viral/bacterial proteins and host proteins.
